Staphylococcus aureus is a Gram-positive, facultative anaerobic bacterium that commonly colonizes human skin and mucous membranes.
To revive the bacteria, thaw the frozen sample on ice or at room temperature, or rehydrate lyophilized bacteria with sterile broth. Using a sterile loop or pipette, transfer a small amount to a suitable growth medium such as Tryptic Soy Agar (TSA) or Mannitol Salt Agar (MSA) and streak for isolation. Incubate at 35–37°C for 18–24 hours under aerobic conditions. After incubation, observe colony morphology, perform Gram staining, and confirm identity with biochemical tests like catalase and coagulase tests. If necessary, subculture to ensure purity before further use.
