WHY MICROBIAL CULTURE MEDIA PREPARATION IS IMPORTANT ???

It is the process of mixing nutrients, agents for buffering and maintaining the osmotic balance, as well as selective inhibitors or indicators to create an agar or broth that supports the growth and the differentiation of microorganisms.

Microbial culture media preparation is a routine task in the regular monitoring of spoilage and pathogenic microbes in microbiological testing.

The media can be of different types
1-Selective and differential Media:
These are media based on either complex or defined media supplemented with growth-promoting or growth-inhibiting additives.
2-Complex Media :
These are rich in nutrients that contain water-soluble extracts of plant or animal tissue (e.g., enzymatically digested animal proteins such as peptone and tryptone)
3-Defined media :
These are the media that composed of pure ingredients in carefully measured concentrations and dissolved in double distilled water.

How a Media is prepared ????
Media preparation in microbiology is the process of mixing nutrients, buffering agents, and other ingredients to create a broth or agar that supports the growth of microorganisms.Steps for preparing media :

-Weigh the ingredients
-Dissolve the ingredients in distilled water
-Adjust the pH if needed
-Add agar and boil to dissolve
-Pour the media into a flask
-Autoclave to sterilize the media at 121°
-Let the media cool slightly
-Pour the media into sterile Petri plates inside the Laminar Air flow
